Monoclonal antibodies are produced by a single clone of cells and are used to target a specific antigen, while polyclonal antibodies are produced by a mix of different clones of cells and are used to target multiple antigens. Antibodies are indispensable research tools that have enabled researchers across diverse scientific fields to specify, identify, locate and/or quantify their protein targets.
Antibodies can be produced through various methods. These include hybridoma technology, phage display, and recombinant DNA technology. Hybridoma technology involves fusing an antibody-producing cell with a cancer cell to produce a hybrid cell that produces the desired antibody. Phage display involves inserting the genetic information for an antibody into a virus and displaying it on the virus surface. Finally, recombinant DNA technology involves cloning the genes for the desired antibody into a vector and expressing it in a host cell. It is a long, frustrating, and potentially expensive process to generate new custom antibodies. The probability of success depends on multiple factors including intended use, biological knowledge of the target’s structure and function, appropriate immunogen design, animal model and clonality chosen, immunization strategy, optimized screening methods, efficient purification, and reliable QC.
