Introduction of Natural Compound Library  

 

For thousands of years, nature has been the source of drugs, and a large number of modern drugs have been separated from nature, many of which are based on their uses in traditional medicine. With the development of compound targets, there is an increasing demand for screening compound diversity. Through the continuous study of natural biodiversity, many of them are still to be developed, and natural products will play a key role in meeting this demand. According to literature reports, in the era of traditional drug research and development, natural products were the most important and almost unique source of new drug research and development. With the development of organic chemistry, medicinal chemistry, computer technology and so on, it is still one of the important means of new drug research and development to transform or design new compounds based on the mother nuclear structure of natural products, even today when synthetic small molecular drugs are dominant.

Among the small molecular drugs that have been approved for marketing, drugs derived from natural products or their derivatives can still occupy half of the country. Natural products are small molecular compounds produced in nature, from any organism, including primary metabolites and secondary metabolites. Natural products have potential biological activities and can be used as lead compounds for drug discovery and commercial development. It is one of the main ideas of developing new drugs based on natural products to optimize their physical and chemical properties (improving solubility, stability, etc.) and biological activities (improving selectivity, reducing toxicity, etc.) by transforming the original structure of natural products or chemically modifying their mother nuclei, so as to obtain a series of natural product derivatives with better medicinal properties. For example, Rosuvastatin is a new type of hypolipidemic drug inspired by the natural extract of Penicillium Mevastatin.

Classification of Natural Compound Library

 

Natural product libraries can be classified according to their structures, such as flavonoids, terpene, alkaloids and phenols.

--Flavonoid Compound Library: Flavonoids are a ubiquitous group of naturally occurring polyphenolic compounds characterized by the flavan nucleus and represent one of the most prevalent classes of compounds in fruits, vegetables and plant-derived beverages. Flavonoids are now known to be ubiquitous in many plants, including the onion (Allium cepa) skin is used as a hay-fever remedy and a digestive anti-inflammatory, and is the active ingredient in a popular OTC nasal spray for sinusitis and allergies. Flavonoids are water-soluble phenolic compounds (having a -OH group attached to an aromatic ring) and are found in the vacuoles of plant cells. Quercetin and rutin are among the most largely found flavonoids in a great variety of fruits and vegetables, including tea, coffee, and other grains.

--Terpene Compound Library: Terpenes are a large and diverse class of organic compounds, produced by a variety of plants, particularly conifers, and by some insects. They are also called isoprenoids—are made up of isoprene molecules. The building block is a five-carbon isoprene (CH2C(CH3)CHCH2) unit . Herbs and higher plants containing their oxidized derivatives, known as terpenoids. Terpenoids are the largest group of phytochemicals consisting of several diverse compounds that are highly diverse in chemical structure, and are a rich reservoir of candidate compounds for drug discovery. Most terpenoids are multicyclic structures that differ from one another not only in functional groups but also in their basic carbon skeletons.

--Alkaloid Compound Library: Alkaloids are groups of natural occurring simple or complex, low molecular weight nitrogen containing compounds which are basic in nature. Alkaloids are secondary metabolites abundantly found in plants, but also to a lesser extent in microorganisms (bacteria and fungi) and animals. In general, a given species contains only a few kinds of alkaloids, though both the opium poppy (Papaver somniferum) and the ergot fungus (Claviceps) each contain about 30 different types. Certain plant families are particularly rich in alkaloids; all plants of the poppy family (Papaveraceae) are thought to contain them. Alkaloids have a wide range of pharmacological activities, including antimalarial, antiasthmatic, anticancer, cholinergic, vasodilator, antiarrhythmic, analgesic, antibacterial and antidiabetic activities.

--Polyphenolic Compound Library: Polyphenols are compounds with various potential biological properties such as antioxidants, anti-inflammatory, antineoplastic, antiaging, cardioprotective, anticancer, and antimicrobial properties. Natural polyphenols play an important role in cancer prevention and treatment by blocking cell cycle, inducing Apoptosis, and inhibiting cell adhesion, migration, proliferation, and differentiation. Polyphenols are defined as compounds having at least one aromatic ring with one or more hydroxyl functional groups attached. Natural polyphenols include flavonoids, phenolic acids, lignans, tannins, stilbenes, curcumin, Coumarin, quinone, and other polyphenols.
